take a look in the admin guide.

use online config with slapd.d.
you can trigger readonly-mode with a simple ldif
on the fly, using the olcReadOnly-Attribute

So I would need to edit the .conf file and restart the service for
this to take effect ?

Is there no "on the fly" solution similar to "FLUSH TABLES WITH READ
LOCK;" for example ?
